Spatial metabolomics-guided deep learning framework for dissecting immune evasion mechanisms in fungal infection microenvironments - Nature

We’re sharing this article early to provide faster access to peer-reviewed, accepted research. It is citable and carries a permanent DOI. This version is subject to further edits and will be replaced automatically by the final Version of Record. All legal disclaimers apply. AbstractInvasive fungal infections cause high mortality in immunocompromised patients, yet the spatially organized metabolic circuits that enable pathogens to evade host immunity remain poorly understood. Here we present an integrated computational–experimental framework coupling high-resolution spatial metabolomics with attention-enhanced deep learning to dissect immune evasion mechanisms within fungal infection microenvironments. We constructed a spatial metabolic atlas from MALDI-MSI and DESI-MSI data acquired on murine invasive pulmonary aspergillosis tissue sections, identifying 1,847 reproducible metabolite features across histologically distinct tissue compartments.
