52-Week Litifilimab Data in Cutaneous Lupus Erythematosus - HCPLive

Response rates with litifilimab, a monoclonal antibody targeting blood dendritic cell antigen 2 (BDCA2), continued to rise through week 52 in patients with cutaneous lupus erythematosus (CLE) refractory or intolerant to antimalarials, according to Victoria P. Werth, MD, professor of dermatology and medicine at the University of Pennsylvania, and François Chasset, MD, PhD, professor of dermatology at Sorbonne Université and Hôpital Tenon in Paris. The 2 discussed the phase 2 AMETHYST Part A data in an HCPLive special report from the European Academy of Dermatology and Venereology (EADV) Congress 2026 in Vienna, Austria (September 30–October 3). The findings come from a Biogen-sponsored congress abstract that has not been peer-reviewed, and both speakers are authors on it.1 AMETHYST Part A (NCT05531565) randomized 93 adults to subcutaneous litifilimab (n = 59) or placebo (n = 34) every 4 weeks through week 24, after which all participants received litifilimab through week 52.1 Chasset noted the cohort was 74% women and 33% non-white.

Ticks don’t quit in the fall: what to check after a brisk walk outside - haligonia.ca

The first frost doesn’t mean you can stop checking for ticks. In Nova Scotia, adult blacklegged ticks, the kind that can carry Lyme disease, are most active in the spring and fall, and the province says they stay active until temperatures are consistently below 4°C. That keeps leaf raking, trail walks and dog walks squarely in tick season this month. Ticks don’t need summer heat, either. The Public Health Agency of Canada says they can be active whenever the temperature is consistently above freezing and the ground isn’t covered by snow, and at any time of year when conditions are right.

Pharmacist Paul Irvine said he understood that women were "concerned" but urged them to speak to their GP or local chemist who could advise them about alternatives. The warning follows official confirmation from the Department of Health (DoH) that supplies of two major HRT patch brands have been disrupted. The affected products include Estradot transdermal and FemSeven Conti, both of which are currently facing manufacturing or distribution delays. Paul Irvine said it was important that women contact their GP or pharmacist for adviceIrvine, a pharmacy manager in Belfast, said much of the time with HRT medications, women often become comfortable with a particular brand. "When they are short and they like to try alternatives, they may suffer side effects or it might just not work as well," he said.

Engineered TYK2 dependence enables myeloid-selective IL-10 receptor agonism - Nature

MainIL-10 is a potent anti-inflammatory cytokine that maintains immune tolerance1,6. Loss-of-function mutations in IL10 or its receptors IL10RA (encoding IL-10Rα) and IL10RB (encoding IL-10Rβ) cause early-onset inflammatory bowel disease (IBD) and autoinflammatory disorders1,7. IL-10 signalling is initiated when IL-10 binds to IL-10Rα and IL-10Rβ to form a tetrameric receptor complex, which is constitutively associated with Janus kinase 1 (JAK1) and tyrosine kinase 2 (TYK2), respectively1,8. This binding triggers JAK1-dependent phosphorylation of IL-10Rα and signal transducer and activator of transcription 3 (STAT3) to suppress pro-inflammatory gene expression and promote immune tolerance8,9. IL-10 primarily exerts its tolerogenic effects in myeloid cells and antigen-presenting cells, such as dendritic cells (DCs) and macrophages, by inhibiting cytokine production, antigen presentation and T cell priming5,10,11,12,13. Loss of IL10RA or STAT3 in these cells leads to colitis and systemic inflammation10,12,13. Despite its central role in immune tolerance, the dual functionality of IL-10 severely limits its therapeutic use6,14.

First 90 days after starting antipsychotics are critical for monitoring diabetes risk, study finds - Medical Xpress

The first 90 days after starting antipsychotic treatment are crucial for changes in blood sugar and weight, finds a new study led by UCL researchers. Second-generation antipsychotics are important treatments for severe mental illness, including schizophrenia and bipolar disorder. However, they can increase the risk of diabetes and weight gain. New research, published in The Lancet Psychiatry, analyzed anonymous NHS GP health records from nearly 97,000 adults in England (ages 18–59) who had severe mental illness and started taking an antipsychotic medicine (olanzapine, quetiapine, risperidone or aripiprazole) between 2000 and 2023.

Cancer is rising in younger adults, and scientists can't fully explain why - Science Daily

Cancer rates are increasing among adults of different ages in England, but a new study has uncovered a particularly concerning pattern among younger people. Rates of bowel and ovarian cancer have risen among younger adults (under 50), while the same increases have not been observed in older age groups. The findings, published in the open-access journal BMJ Oncology, also show that several other cancers are becoming more common at a faster pace among younger adults than among those aged 50 and older.
