How Insertion Sequence-Mediated Gene Amplification Helps Acinetobacter baumannii Adapt to the Host KNOXVILLE, TN, October 18, 2026 /24-7PressRelease/ -- Acinetobacter baumannii is a major nosocomial pathogen with a remarkable capacity to adapt to both hospital and host environments. In this study, the researchers identified ISAba1-mediated amplification of the crp-osmC gene cluster as a previously unrecognized mechanism promoting host-associated fitness. Increased crp-osmC copy number enhanced bacterial growth, competitive fitness, serum survival, and colonization in mouse models. Transcriptomic analyses further linked this amplification to changes in metabolic flexibility and nutrient acquisition. Interestingly, the fitness advantage was accompanied by reduced biofilm formation and increased susceptibility to several antibiotics, revealing a potential evolutionary trade-off between host adaptation and antibiotic resistance in A. baumannii.
This study was led by Yuqin Song and Jie Feng from the Institute of Microbiology, Chinese Academy of Sciences.

Rational engineering of Omp34 loop-based immunogen enhances protection against Colistin-resistant Acinetobacter baumannii - Nature

AbstractThe surface antigens are promising immunogenic targets for vaccine development and immunotherapies. Here, the loop3 to loop7 (L3-L7) region of the Omp34 from Acinetobacter baumannii was evaluated. A DNA fragment encoding the L3-L7 region was amplified from the Omp34 gene. The structures of ΔL5/5Gly-L3-L7 and ΔL5/Acr-L3-L7 were achieved through the modification of the loop 5 sequence in the L3-L7 of Omp34. Optimization of Omp34 loop epitopes via Acr sequence linkage was performed. Omp34, L3-L7 region, ΔL5/5Gly-L3-L7 and ΔL5/Acr-L3-L7 were recombinantly expressed. Mice immunized with the purified antigens, were infected with the clinical isolate of A. baumannii 58ST. Active immunizations with ΔL5/Acr-L3-L7, L3-L7, Omp34 and ΔL5/5Gly-L3-L7 resulted in 75%, 50%, 37.5% and 37.5% survival rates, respectively.

Chaperone-mediated autophagy curbs senescence - Nature
Sereda and colleagues show that chaperone-mediated autophagy declines with aging, which leads to enhanced senescent cell burden that is driven, in part, by failed macrophage-mediated immunosurveillance that can promote tissue dysfunction and systemic aging.
